? 首次證實了在序列和結構上高度復雜的Piezo蛋白是在哺乳動物中鑒定發(fā)現的首類機械門控陽離子通道的孔道蛋白,從而確立了機械門控Piezo通道這一全新離子通道家族類型[Nature(Article)2012; Nature(Article)2015; Neuron 2016],開辟了神經科學、生理學、藥理學、細胞生物學以及離子通道等研究領域新的研究方向。
? 深入揭示了Piezo通道將機械力刺激高效轉化為電化學信號的獨特結構基礎和精巧工作機制,并鑒定發(fā)現了其小分子藥物、相互作用蛋白以及剪切變體調控機制[Nature (Article)2018; Nature(Article)2019; Neuron 2020; Nature Communications 2017; Nature Communications 2018]。課題組創(chuàng)新性的開展了先結構后功能的離子通道研究范式,與合作者在國際上首次解析并報導了鼠源Piezo1、Piezo2以及由其課題組所鑒定發(fā)現的Piezo1剪切變體Piezo1.1的冷凍電鏡三維結構,揭示了其包含共計114次跨膜螺旋區(qū)的三聚體螺旋槳狀結構,進而結合電生理等功能研究手段揭示了其離子通透與門控機制,并系統(tǒng)性提出了Piezo通道進行機械門控的功能區(qū)模塊化作用機制假說、杠桿作用機制假說、雙門控作用機制假說、以及門塞和閂鎖作用機制假說,有力推動了對Piezo通道這類全新復雜離子通道家族的理解。
